I just finished cutting all my 2x4's for my enclosure and want to make the frame, but im not sure if im supposed to put the sealant on before or after it's built.

Are the 2x4's going to be internal or external supports? If they're external you don't really need to seal them. As a whole you generally seal the entire surface (all sides) of any wall face within the enclosure. This helps with wood swelling. Anything external to that would not require any particular sealing.

Though I believe more to the point - it would be easier to cut your pieces to their correct lengths, do a test fit, and then seal them individually.
